The Three Sisters route starts in Medina, Texas, located 63 miles (101 km) North West of San Antonio. The Three Sisters comprise of Ranch Road 335, Ranch Road 336, and Ranch Road 337 all of them two-lane roads with some stretches of bumpy road. This route is full of twisties through Texas, so twisty that the Three Sisters are often referred to as the "Three Twisted Sisters". The ride will take under 4 hours in 132 miles (213 km).

From Medina ride onto Ranch Road 337 (RR337) heading West. Take RR337 until you come up on the town of Leakey approximately 37 miles (59 km) into the route. RR337 is one of the Three Twisted Sisters, she is the longest and twistiest of the three! Passing through extreme natural beauty of canyon land in Texas Hill Country, RR337 is renowned for its hairpin turns and switchbacks, it has been called one of the most scenic drives in Texas and was ranked number 16 on Texas Monthly's April 2006 Issue "Top 75 Things We Love About Texas" http://esr.cc/NsM6mv

From Medina the first stop is Leakey, a small town of 387 people. It's a frequent stop for riders on RR337, and the locals have taken advantages of the Three Twisted Sisters notoriety by selling souvenirs at Frio Canyon Motorcycle Stop. Leaving Leakey, go North on US Route 83 for about a mile and look for Ranch Road 336 (RR336) on the left. Continue North on RR336 until you meet Texas Route 41. This portion of the route is 27 miles (44km) of twisty road though Texas Hill Country.

From the RR336 and Route 41 Intersection, go west on Texas Route 41 for 14 miles (22 km) then take a left heading South to get onto Ranch Road 335 (RR335) to Camp Wood, a small town of 822 people. Here you can gas up, or eat and sleep at the few local diners and motels. Check out Nueces River for swimming, fishing, kayaking, canoeing, camping, birding, and hiking activities to be enjoyed in the Camp Wood area. Leave Camp Wood and head East back on to RR337 for some more twisties and finish the route when you reach Leakey a second time.

From Medina to Leakey to Camp Wood and then back to Leakey for chock full of twisties through Texas Hill Country, so twisty that the roads are referred to as the Three Twisted Sisters.
